The aim of this study was to evaluate transcriptional activity of PLA2G7 (platelet-activating factor acetylhydrolase) and ACE (аngiotensin-сonverting enzyme) genes in patients with a history of anaphylaxis. Methods. 97 children were enrolled in the study: 27 children with a history of food anaphylaxis; 35 children with atopic dermatitis; 35 children without allergic diseases: 26 children were IgE-negative, 9 children were IgE-positive («Phadia 100»; «Phadia AB», Sweden). RNA was extracted from blood samples with Trizol RNA Prep kits («Isogen», Russia). cDNA production was performed with MMLV RT kit («Evrogen», Russia). Real-time PCR was held with CFX-96 device («Bio-Rad», USA) and qPCRmix-HS PCR mix («Evrogen», Russia). Oligonucleotide primers and probes were synthesized at «DNK-Sintez» (Russia). Results. In children with a history of food anaphylaxis decrease in PLA2G7 gene expression was detected comparing to control groups (p&lt;0,05). Transcriptional activity of PLA2G7 and detection rate of ACE gene expression were lower in children with moderate/severe anaphylaxis comparing with mild anaphylaxis children (p&lt;0,05). The study revealed relationship of PLA2G7 and ACE gene expression and clinical features of anaphylaxis. Conclusion. Reduction of blood cells PLA2G7 and ACE mRNA levels in children with a history of food anaphylaxis may be considered to be a biomarker of severe anaphylaxis.</p></abstract><trans-abstract xml:lang="ru"><p>Цель. Выявить особенности экспрессии генов PLA2G7 и ACE у пациентов с пищевой анафилаксией (вне анафилактической реакции). Материалы и методы. В исследование включены 27 детей с диагнозом «пищевая анафилаксия», группу сравнения составили 35 детей с атопическим дерматитом. В группу контроля вошли 35 детей без аллергических заболеваний, которые были распределены на 2 подгруппы: К1 - с отсутствием сенсибилизации (n=26), К2 - с латентной сенсибилизацией (n=9). Выделение РНК из цельной крови осуществлялось с использованием наборов Trizol RNA Prep (ООО «Лаборатория Изоген», г. Москва). Для получения кДНК использовался набор MMLV RT kit (ЗАО «Евроген», г. Москва). ПЦР в режиме реального времени проводилось на приборе CFX-96 (Bio-Rad, USA) с использованием смеси для ПЦР qPCRmix-HS (ЗАО «Евроген», г. Москва). Праймеры и зонды синтезированы ООО «ДИК-синтез» (г. Москва). Результаты. У пациентов с анафилаксией выявлено снижение экспрессии гена PLA2G7 относительно детей групп контроля (p&lt;0,05). Показано снижение экспрессии гена PLA2G7 и частоты обнаружения экспрессии гена ACE у пациентов с анафилаксией средней и тяжелой степени тяжести относительно детей с анафилаксией легкой степени (р&lt;0,05). В ходе исследования выявлена взаимосвязь экспрессии генов PLA2G7 и ACE с клинической картиной анафилаксии. Заключение.
