The risk of systemic side effects during allergen-specific immunotherapy sets the task for researchers to create modern therapeutic forms of allergens with high immunogenic and low allergenic activity. Our laboratory has been developing preparations for the diagnosis and treatment of house dust mites allergies for many years.</p> <p><bold>AIMS: </bold>To create modern therapeutic forms of allergen preparations from house dust mites of the genus <italic>Dermatophagoides</italic> that are intended for allergen-specific immunotherapy</p> <p><bold>MATERIALS AND METHODS: </bold>Physicochemical and immunobiological methods, such as electrophoresis in polyacrylamide gel, micropoint immunoblot, solid-phase enzyme-linked immunosorbent assay, and the reaction of binding inhibition of allergen-specific immunoglobulin E in the sera of patients were used to study the obtained preparations.</p> <p><bold>RESULTS:</bold> A technology has been developed to obtain a granular dosage form of a mixed allergen from <italic>Dermatophagoides pteronyssinus</italic> and <italic>Dermatophagoides farinae </italic>mites for sublingual use. This treatment form has successfully passed preclinical tests, has a pronounced immunogenic activity, and reduced allergenicity, and is convenient for pediatric practice. Allergoids, which are chemically modified preparations, are obtained to reduce allergenicity and increase therapeutic allergen immunogenicity. A succinylated monomeric house dust mites allergoid <italic>Dermatophagoides pteronyssinus</italic> was created and studied with the National Research Center, Institute of Immunology Federal Medical-Biological Agency of Russia. The study of the immunobiological properties of the obtained preparation showed that the monomeric allergoid has increased immunogenic and decreased allergenic activity contrary to the native water-salt extract.</p> <p><bold>CONCLUSIONS:</bold> The created forms of mite allergens can be used to treat patients who are sensitized to house dust mites of the genus <italic>Dermatophagoides</italic> after clinical trials.</p></abstract><trans-abstract xml:lang="ru"><p><bold>ОБОСНОВАНИЕ.</bold> Рост аллергических заболеваний диктует необходимость разработки новых форм лечебных аллергенов, так как терапия нативными водно-солевыми экстрактами аллергенов не является безопасной и удобной. Риск системных побочных эффектов при проведении аллергенспецифической иммунотерапии (АСИТ) ставит перед исследователями задачу создания современных лечебных форм аллергенов, обладающих высокой иммуногенной и низкой аллергенной активностью. Наша лаборатория в течение многих лет занимается разработкой препаратов для диагностики и лечения клещевой аллергии.</p> <p><bold>ЦЕЛЬ</bold> ― создание современных лечебных форм препаратов аллергенов из клещей домашней пыли рода Dermatophagoides, предназначенных для АСИТ.</p> <p><bold>МАТЕРИАЛЫ И МЕТОДЫ.</bold> Для исследования полученных препаратов применяли физико-химические и иммунобиологические методы: электрофорез в полиакриламидном геле; микроточечный иммуноблот; твердофазный иммуноферментный анализ; реакцию торможения связывания аллергенспецифического IgE в сыворотках крови пациентов.</p> <p><bold>РЕЗУЛЬТАТЫ.</bold> Разработана технология получения гранулированной лекарственной формы микст-аллергена из клещей <italic>Dermatophagoides pteronyssinus</italic> и <italic>Dermatophagoides farinaе</italic>, предназначенной для сублингвального использования. Данная лекарственная форма успешно прошла доклинические испытания, обладает выраженной иммуногенной активностью, сниженной аллергенностью и удобна для применения в педиатрической практике. Для снижения аллергенности и увеличения иммуногенности лечебных аллергенов получают химически модифицированные препараты ― аллергоиды. Совместно с ФГБУ ГНЦ «Институт иммунологии» ФМБА России был создан и изучен сукцинилированный мономерный клещевой аллергоид <italic>Dermatophagoides pteronyssinus</italic>.
